The objective of this study was to identify the cellular antioxidant effects of chitin oligosaccharides (NA-COS), produced by hydrolysis of crab chitin in 12N hydrochloride acid for 7 hours at 40 0C. Their chemical structure was determined using FT-IR, NMR spectrometry. TOF-Mass spectrometry was used to determine their molecular weight (Mw). We investigated the inhibitory effect of NA- COS on myeloperoxidase (MPO) activity in human myeloid cell (HL60),
and its inhibition of oxidative DNA damage. Furthermore, direct radical scavenging effect was determined in human fibro-sarcoma cell (HT1080) using the 2’,7’-dichlorfluorescein-diacetate (DCFH-DA) fluorescence probe. NA-COS with Mw 229.21- 593.12 Da exhibited cellular radical scavenging activity at 1μg/ml assessed by the increase DCF fluorescence intensity with incubation time and its MPO activity. In addition, it was found that NA-COS inhibited the
oxidative DNA damage of cell. These results indicate that NA-COS could act as a potent radical scavenger in live cells.
