Identification of 3-methoxy-4-hydroxybenzoic acid and 4-hydroxybenzoic acid with Antioxidative and Antimicrobial Activity from Arachis hypogaea Shell

The methanol extract of Arachis hypogaea shell showed antioxidative and antimicrobial activity. The methanol extract was successively purified by solvent fractionation, silica gel adsorption column chromatography, Sephadex LH-20 column chromatography and octadecylsilane column chromatography. The purified active substances were isolated by high performance liquid chromatography, and were identified as 3-methoxy-4-hydroxybenzoic acid and 4-hydroxybenzoic acid by LC-MS and GC-MS. The amount of 3-methoxy-4-hydroxybenzoic acid and 4-hydroxybenzoic acid were 3.8 mg and 9.8 mg per kg of shell, respectively.

땅콩(Arachis hypogaea)껍질의 MeOH 추출물은 항균 및 항산화 활성을 보여 땅콩껍질에 함유된 활성물질의 탐색을 시도하였다. MeOH 추출물을 solvent fractionation, silica gel adsorption column chromatography, Sephadex LH-20 column chromatography, ODS column chromatography 등으로 정제하고 HPLC에 의해 활성물질을 분리하였다. 분리된 두 활성물질은 LC-MS, GC-MS분석에 의해 4-hydroxybenzoic acid와 3-methoxy-4-hydroxybenzoic acid로 동정되었으며 두 물질은 땅콩껍질에 각각 3.96 mg/kg, 9.73 mg/kg정도 함유되어 있었다.
