Fed-batch cultivation of transgenic rice cells for the production of hCTLA4Ig using concentrated amino acid

Typical expression characteristics of RAmy3D promoter in transgenic rice cell suspension cultures is that target protein can be expressed inducibly in sugar depletion condition1,2). When we use this promoter, growing media of batch culture should be exchanged with production inducible sugar free medium for the production of target protein, human cytotoxic T-lymphocyte antigen 4-immunoglobulin (hCTLA4Ig). In this study, after depletion of sugar in growth media, a newly-developed fed-batch culture method was applied by feeding only concentrated amino acid and we can increase the production yield of target protein by 2.7 fold than the batch culture without media exchange with production inducible sugar-free media. This fed batch culture was
performed in a 15-L stirred-tank bioreactor and it was found that maximum hCTLA4Ig production level was 171.56 mg/L in 12 days (texpression) by feeding concentrated amino acid only.
