TGE(Transmissible Gastroenteritis) is caused by a virus belonging to coronavirus, that causes an acute infection of the small intestine of the pig. No drugs are effective against TGE virus in swine. Thus, the development of TGE virus vaccine is keenly needed. TGE virus vaccine can be produced efficiently by employing ST-1 cells in animal cell bioreactor. Glutamine has been known to be a major energy source for mammalian cells. In this study, it was confirmed that TGE virus vaccine productivity was improved by 2.7 times with fed-batch addition of glutamine. However, the addition of glutamine resulted in the accumulation of ammonium ion, toxic waste metabolite. To overcome the dilemma of glutamine and ammonium ion, an novel approach was tried. The ammonium ion was removed from medium by using zeolite while providing
additional glutamine, and the kinetics of cell growth and virus titer were monitored. It was confirmed that vaccine productivity was improved by three times in a novel culture process which combined fed-batch feeding of glutamine and removal of ammonium ion using zeolite.
