Enhanced Production of Monacolin-K through Medium Optimization by Response Surface Methodology

초록

영어

In this study, we intended to increase monacolin-K production through medium optimization by statistical method. Solid state fermentation in flasks was undertaken to learn how different compounds in the fermentation medium affect the monacolin-K production. Monacolin-K production by basic production medium in solid state flasks was 0.130 mg/g. It was found that monacolin-K production by Monascus pilosus are influenced by several factors such as
glucose, peptone water, dipotassium phosphate, sodium nitrate and L-alanine. Plackett-Burman experimental design was used to determine the key factors which are critical to produce the biological products in the fermentation. According to the result of Plackett-Burman experimental design, a second order response surface design was applied using initial water content, substrate(rice), glucose, and dipotassium phosphate as factors. Applying the response surface
methodology(RSM), the optimum concentration of the four variables was obtained. The maximum monacolin-K production(1.44 mg/g) predicted by RSM agrees well with the experimental value(1.23 mg/g) obtained from the experimental verification at the optimal medium.
