To improve the production of H. chejuensis red pigment, analyzed as prodigiosin, mutant strain M3349 was developed using by mutagenesis. To investigate the bioprocess parameters significantly influencing prodigiosin production of mutant strain M3349, tow-level Plackett-Burman design was used. Among fourteen factors tested, NaCl, Na2SiO3, MgCl2, H3BO3, Na2HPO4, Na2SO4, CaCl2 were selected, which revealed high significant effect on prodigiosin production. Optimum medium formulation was obtained using the three-level Box-Behnken design. The optimal combination of medium evaluated from the non-linear optimization algorithm of statistical experimental designs was as follows : sucrose 10 g/ℓ, peptone 8 g/ℓ, yeast extract 2 g/ℓ, NaCl
10 g/ℓ, H3BO3 0.022 g/ℓ, Na2SiO3 0.008 g/ℓ, Na2SO4 12 g/ℓ, MgCl2 0.7 g/ℓ, Na2HPO4 0.02 g/ℓ and CaCl2 1.8 g/ℓ. Prodigiosin production by wild type using basal medium was 0.5 g/ℓ. But prodigiosin production of mutant strain M3349 by optimal combination was 2.6 g/ℓ, which was about 5 times higher than wild type using basal medium.
