In this work, we have cultivated a few Bacillus species. Screened from soil microorganisms in shaking flask. The Bacillus sp. BKS 14, 15 and 39 produced the enzyme catalase. Cell density was determined by measuring the optical density at 600 nm. Spectrophotometer was used for
determination of catalase activity. Effects of temperature and pH on the catalase activity were investigated in the range of 30 ~ 70 ℃ and pH 3 ~ pH 13.

토양시료에서 얻은 각기 다른 콜로니로부터 카탈라제 생산균주를 1차 선별하고 효소 활성이 가장 높은 3가지 다른 바실러스를 본 연구에 사용하였다. 각각의 균주는 TSB 복합배지, 30 ℃, 200 rpm에서 배양하여 시료를 채취하여 원심분리하고, 펠릿은 제거된 상등액을 수집하여 효소 활성을 측정하였다. 배양하여 얻어낸 상등액에 과산화수소를 기질로 사용하여 반응시킨 후, 발색시약을 넣고 30분 후 염산으로
반응을 종결한 후 분광광도계를 이용하여 525 nm에서 흡광도를 측정하였다. 뿐만아니라 얻어진 효소의 온도와 pH 변화에 따른 효소 활성 변화를 측정하였다.
