CLONING AND CHARACTERIZATION OF ALGINATE LYASE FROM STREPTOMYCES SP. ALG-5

An alginate lyase-degrading marine bacterium, Streptomyces sp. ALG-5, was isolated from seaweed using M9 alginate agar plate. We have investigated the optimum condition for the cell growth and alginate lyase production from Streptomyces sp. ALG-5. The effects of temperature, pH and metal ions on the extracellular alginate lyase activity were investigated. The gene of alginate lyase of Streptomyces sp. ALG-5 was cloned by PCR with specific primer designed
from homologous nucleotide. The alginate lyase gene was successfully expressed in Escherichia coli BL21. The recombinant cells exhibit a high alginate lyase activity.
