A microorganism utilizing rice hull and rice bran was isolated from sea-water in Korea and identified by 16S rDNA sequencing and partial sequencing of gyrase A coding DNA as Bacillus velezensis. Production of cellulase by this strain, named as B. velezensis A-68, was examined in the liquid medium containing 2.0%(w/v) glucose, 0.25% yeast extract, 0.5% K2HPO4, 0.1% NaCl, 0.02% MgSO47H2O and 0.06% (NH4)2SO4 at 30℃ for 72 hr under aerobic conditions.
The best carbon source and nitrogen sources for the production of cellulose by B. velezensis A-68 were found to be rice hull and yeast extract. The highest production of cellulase was obtained when the concentrations of rice hull and yeast extract were 3.0% (w/v) and 0.25% (w/v), respectively. Optimal initial pH of the medium and temperature for production of cellulase were 7.3 and 40℃. The gene coding for cellulase produced by Bacillus velezensis A-68 was
isolated and coned. The deduced amino acid sequence of the cellulase showed high identity to cellulases from other Bacillus species.
