Various induction methods, including chemicals addition and physical methods, have been reported and used for protein production in Escherichia coli. Among these, oxygen limitation is the simple and economically feasible induction method for protein production by E. coli because this method does not need any inducer, and reduces the energy input required for aeration and agitation in the fermentation process. The oxygen-sensitive promoter, a modified nar promoter, is
maximally induced under microaerobic conditions, and can be used for expression of -galactosidase in the absence of nitrate β ion as previously reported. This modified nar promoter has higher strength for the anaerobic protein synthesis than those previously reported. However, this promoter did not overexpress other type of proteins, and there is no report about the expression of other economically useful proteins except for β-galactosidase. So, we modified
the promoter to be able to produce the various proteins by the modified nar promoter. Our result shows the possibility of this modified nar promoter system as a versatile tool for economical production of recombinant proteins in E. coli.
