Sialic acid (N-acetylneuraminic acid) is an interesting and important molecule involved
in glycosylation of eukaryotic proteins and bacterial cell wall synthesis. Sialyloligosaccharides consisting of sialic acids are known to exhibit beneficial effects on microbial flora in human intestine as a prebiotics. In order to synthesize sialyloligosaccharides using enzymatic process, CMP-sialic acid is required as a sugar donor. However conventional E. coli strains including E. coli K12 strain cannot synthesize CMP-sialic acid because two key enzymes are missing. We introduced the missing metabolic reactions catalyzed by sialic acid synthase (NeuB) and CMP-sialic acid synthase (NeuA) in a genome scale metabolic model of E. coli K12 strain for the
evaluation of feasibility of CMP-sialic acid production in recombinant E. coli K12 strain. Maximum theoretical yield of CMP-sialic acid from glucose was predicted to be 0.263 mol CMP-sialic acid/mol glucose. Also, the effect of gene knock-out on CMP-sialic acid production was investigated through stoichiometric modeling.
