Thioredoxin-1 (TRX) is a redox-active protein with multiple intracellular and extracellular functions. Thioredoxin is released from cells in response to oxidative stress and high TRX levels in plasma or serum are good markers for oxidative stress associated with cancer. We measured LPS-induced cytotoxicity by MTS assay, intracellular ROI levels by DCFH-DA and detected
over-production of the nitric oxide by Griess’ reagent in melanoma A375 cells. Exogenous TRX prevented inflammatory response and inhibited ROI and NO production. We used His-TRX pull down experiment, immunoprecipitation, LCMS/ MS, and ESI Q-TOF MS/MS to identify membrane proteins as a binding partner for TRX entry protein. As a result, several proteins expressed on the plasma membrane were found to bind exogenous TRX, which may be important for TRX internalization and signal transduction. Studies are in progress to
understand the molecular mechanisms of the TRX entry under the inflammatory response and oxidative stresses.
