Aging Affects Expression of HSP60 in mice

Background : Heat-shock protein is a family of highly-conserved protein. HSPs are believed that they are important in protection of cells from harmful stimuli.1) HSP60, one of the major families of HSPs, is generated in response to tissue injury, and/or stress.2) Aging is the accumulation of changes responsible for the sequential alteration.3) Many of the theories about molecular aging imply that age-dependent changes occur in the molecular configuration of expression of
proteins.4) We examined the effect of aging on HSP60 expression in variable tissues. Method : We used ICR mouse and divided 3 groups; 50days, 3months, and 8months old. Heart, lung, liver, kidney, and skin tissues were acquired from each groups. Proteins were extracted from each tissue, and analyzed using western blot. Result and discussion : The synthesis of HSP60 was similar in skin tissue with aging. In lung tissue, HSP60 was more expressed in young mice.
In heart, liver, and kidney, we obtained two different bands. Smaller HSP60(lower band) increased with aging. Difference of size was due to signal sequence, 26 amino acid residues at the N terminus. Expression pattern of HSP60 was different in each tissues and each aging groups.
