Azoreductases (EC 1.6.5.2) is an enzyme mediating reduction of azo-dye depending on specifically either NADH or NADPH. To understand the physiological response of reducing power perturbation, Escherichia coli strains expressing Enterococcus faecalis azoreductase (NADH-dependnet, EFAR) and Staphylococcus aureus azoreductase (NADPH-dependent, SAAR) were constructed as a model. The induced enzyme activities of the cell extracts harboring EFAR and SAAR were 7.1 and 3.0 µM/mg-protein, respectively. The growth of E. coli under LB-medium with methyl red substrate was 78% of control when EFAR was expressed while 58% when SAAR was expressed. The results indicated the intracellular NAD(P)H pool perturbation influenced the cellular growth. We are currently further studying on the cellular regulatory response according to the reducing power perturbation by transcriptom and proteom analysis as well as the variation of intracellular NAD(P)H pool.
