The response to phosphate starvation by PhoR-PhoB two-component regulatory system is controlled by other regulatory systems in Escherichia coli. In this study, the interactions among PhoB, PhoR, PhoU and CreC were investigated at combined transcript and protein levels using real-time PCR and fluorescence resonance energy transfer (FRET) analyses. The expressions of the phoB, phoR and creC genes are regulated depending on the concentration of environmental phosphate. The protein-protein interactions between PhoB-PhoR and -CreC are occurred depending on the concentration of environmental phosphate. These suggest that PhoU is a no direct regulator but CreC is an alternative regulator. These results showed that the detailed time- and phosphate concentration-dependent interactions among PhoB, PhoR, PhoU and CreC in response to phosphate starvation of E. coli.
