The production of doxorubicin derivative in an engineered strain of Streptomyces venezuelae

In order to develop a clean host for combinatorial biosynthesis of glycosylated macrolides, an engineered strain of Streptomyces venezuelae was constructed by deletion of desosamine biosynthetic cluster as well as pikromycin polyketide synthases (Pik PKSs). The engineered deoxysugar pathways including TDP-L-daunosamine was introduced to the mutant strain. The ε-rhodomycinone were fed to the growing medium of those mutants, successfully producing their
daunosaminyl derivatives. These results demonstrate a successful attempt of creating structurally diverse hybrid macrolides using S. venezuelae system as well as provide further insight into the substrate flexibility of glycosyltransferases together with their auxiliary proteins.
