The isoflavone-O-β-D-Glucan extract is a potent chemical component in the extract from the submerged-liquid culture of Agaricus blazei Murill. In the present study, the anti-tumor effect of isoflavone-O-β-D-Glucan extract was investigated using MCF-7 cells. MCF-7 cells were maintained with Dulbecco's Modified Eagle Medium/Ham's F-12 nutrient (DMEM/F12) supplemented with 10% fetal bovine serum (FBS) at 37℃ in a humidified 5% CO2. For cell
proliferation experiments, cells were seeded in 90 ㎜ dishes, treated with the various concentrations (0, 1, 5, 10, 15, 20, 25 ㎕) of the isoflavone-O-β-D-Glucan extract (10 ㎎/㎖) for over a period of 96 hr of incubation. The isoflavone-O-β-D-glucan inhibited the proliferation of cells in a dose-and time-dependent manner, which was mediated through up-regulation of
p21/p53 and down-regulation of cyclin D. The results suggest that isoflavone-O-β-D-Glucan extract inhibited the proliferation of MCF-7 cells by induces apoptosis and block cell cycle progression in the G/S phase. This study was partly supported by the BK21 program of GNU.
