Enhancement of isoflavone contents in soybean by soaking and germination was developed, and fermentation conditions of Chungkukjang were optimized by using response surface methodology analysis. For high-yield isoflavone production, soaking conditions were at 25.6℃, 7 hour and gemination conditions were 29.1℃, 42.4 hour, respectively. Total isoflavone contents in soybean by treatment was increased from 1112 ppm to 1900 ppm. Also, optimum fermentation conditions of Chungkukjang were determined 39.96℃, 1.32%(inoculumn size) and 42.4 hour, respectively. Isoflavone contents under optimum fermentation conditions of Chungkugjang was 1.3 times higher than control(1960 ppm).
