Elastin-like polypeptide (ELP) is a recombinant polypeptide composed of repetitive pentapeptide Val-Pro-Gly-Xaa-Gly. which undergoes an inverse phase transition by thermal stimuli1. This polypeptide is soluble in aqueous solution below a characteristic inverse transition temperature (Tt) (hydrophilic state) but aggregates above Tt (hydrophobic state). The phase transition is reversible and occurs very rapidly within ~2 ℃. ELP is expected to be a prospective biomaterial due to its low endotoxicity, cytotoxicity and no antigenic properties as well as the stimuli-responsiveness2. ELPs with a variety of lengths were synthesized by recursive directional ligation method (RDL) and simply purified by inverse phase transition cycling3. We investigated the phase transition of ELPs by varying several factors including polypeptide length, ion
strength and solvent using a UV spectrometer. In the presentation, the potential applications of the designed ELPs will be demonstrated as well.
