In an attempt to search for new therapeutic materials, the anti-allergic effect of Ecklonia cava (E. cava) was observed as a part of a screening study carried out to identify biologically active antiallergic medicinal sources from number of marine algal species. In this study, we investigated the inhibitory effect of E. cava extract on the expression of the FcεRI in human basophilic KU812F cells. Flow cytometric analysis revealed that E. cava extract exerts a
concentration-dependent reduction in expression of FcεRI on cell surface. Latter experiments further revealed that this extract is capable of reducing the binding between human myeloma IgE, or mite allergen-specific IgE, and cell surface FcεRI. Interestingly, E. cava extract reduced total cellular FcεRI α chain mRNA expression analyzed using RT-PCR. Moreover, data obtained using fluorescence spectrophotometer revealed that E. cava extract also inhibits FcεRI-mediated
histamine release in aconcentration-dependent manner. These results suggested that E. cava extract negatively regulates FcεRI expression and basophil degranulation and could be a potential candidate to carry out further studies concerning about protection from allergic diseases.
