Inhibitory Effect of Methanolic Extract from Dictyopteris prolifera on Production of Reactive Oxygen Sspecies and Matrixmetalloproteinase-2 and -9 Activities

Recently, Reactive Oxygen Species (ROS) have been studied extensively, because ROS cause not only aging but also numerous diseases. The aim of this study, therefore, is to study ROS scavenging effect from a seaweed, Dictyopteris prolifera(DP) which is a kind of brown algae, and inhabit in the lower part of littoral zone and subtidal zone. Because little is known of the
efficacy of the DP, we investigated the effect of DP extract on scavenging effect of ROS. Direct radical scavenging effect were carried out with the measurement of electron spin resonance (ESR) and dichlorofluorecin diacetate (DCFH-DA)fluorescence assay in human fibrosarcoma cell line (HT1080). DP extract exerted a potent radical scavenging activity. Besides, the DP extract
inhibited myeloperoxidase (MPO) activity which coverts H2O2 into HOCl in human myeloid cell (HL60) and DNA oxidation caused by hydroxyl radical generated by Fenton reaction. These experiments suggested that DP can inhibit the generation of ROS in dose-dependant manner. As well, it is known that ROS is related with matrixmetalloproteinase (MMP) expression, so we also
investigated MMP-2 and -9 activity using HT1080 cell. The result showed the inhibitory effect of DP extract on MMP-2 and MMP-9 activitiesand similar trend with results of ROS scavenging. The more ROS is eliminated, the more expression of MMP-2 and -9 is suppressed. Above results suggest that DP extracts inhibits can MMP activity via scavenging activity of ROS.
