Menaquinone-8 pathway of Escherichia coli was metabolically engineered by modulating the
expression levels of two precursor pathways: isoprenoid and 1,4-dihydroxy-2-naphthoate pathways.
Overexpression of E. coli isoprenoid pathway genes (dxs, dxr, ispA, IspB and idi) was found to
have significant effects on menaquinone-8 formation. Among them, the overexpression of idi
showed most significant effect on menaquinone-8 production followed by ispA and dxr. Similar
positive effect was also observed when 1,4-dihydroxy-2-naphthoate pathway genes (ubiE, menB, menC, menD, menE, menF, and menA) were overexpressed. The overexpressions of menA and menB were most effective in menaquinone-8 production. This result suggests that precursor pool engineering will be a good strategy for menequinone overproduction in E. coli.
