Ceramide is important not only for the maintenance of the barrier function of the skin but also for the water-binding capacity of the stratum corneum. Though the effectiveness of ceramide is not understood fully, ceramide has become a widely used ingredient in cosmetic and pharmaceutical industries.1) Compared to other sources of materials, yeast is more suitable for the production of ceramide because yeast grow fast and are non-toxic. However, ceramide production from yeast has not been widely studied and most work was carried out for only two strains: Saccharomyces cerevisiae and Torulopsis (Candida) utilis and the quantity are very low.2) Regulation of ceramide synthesis components are required for the increasing of ceramide production. In this study we investigated the effect of genes, lcb1, lcb2, tsc10, lac1, lag1, and
sur2. The genes were cloned on pESC-URA vectors. S.cerevisiae was cultivated in YPDG medium at 30℃. Ceramide was prepared by the cell extracts by solvent extraction and analyzed by High Performance Liquid Chromatography using Evaporative Light Scattering Detecctor.
