Overexpression of Non-mevalonate Pathway in Escherichia coli to Increase Isoprenoid Pool for Terpenoids Production

Isopentenyl diphosphate (IPP) and its isomer dimethylallyl diphosphate (DMAPP) constitute the basic building block of isoprenoids, a family of compounds that has diverse structure and
function. The distribution between glyceraldehyde 3-phosphate (G3P) and pyruvate, the originating precursors of the isoprenoids in MEP pathway, is a major factor that can limit isoprenoid production yields in E.coli. We have overexpressed the E.coli D-1-deoxyxylulose 5-phosphate synthase (DXS) to increase deoxyxylulose 5-phosphate (DXP) and, consequently increase the isoprenoid pool by overexpressing isopentenyl diphosphate isomerase (IDI) to overproduce the isoprenoid precursor in Escherichia coli. Engineered host can be applied for the
production of various classes of terpenoids.
