This study was conducted to establish the optimal concentrations of growth hormones and the suitable explants among leaf and stem on callus induction and plant regeneration of bitter gourd (M omordica charantia L.) Different explant cultured on MS medium with various combinations of BAP(0-4 mg/L), NAA(0-1 mg/L), Kinetin(0-2 mg/L), Zeatin(0-2 mg/L) and 2,4-D(0-1 mg/L). Callus induction from stem and leaf explant were observed after 1 weeks of culture.
The highest callus induction frequency from stem explant was obtained with 1 mg/L 2,4-D and 4 mg/L BAP. To analyze the effect of leaf age along callus formation, we measured number of induced callus, induction rate, fresh and dry weight of leaf explant. The frequence of callus induction was higher in the young leaf explants then the older ones. After transfer of embryogenic callus on plant regeneration medium, shoots were induced. The regenerated shoots were transferred in 1/2 MS medium with 0.5 mg/L NAA for root formation.
