Bone marrow-derived mesenchymal stem cells (BM-MSCs) have been used in tissue engineering and regenerative medicine because of their multipotency. For therapeutic use of BM-MSCs, large quantities of cells are required. However, BM-MSCs lose the ability to proliferate and maintain phenotypes while they are cultured. BM-MSCs can be expanded on microcarriers in spinner flask, which provide large surface area. Expanded BM-MSCs in spinner culture system can provide the possibility of obtaining sufficient number of cells with maintaining their phenotypes. In this study, BM-MSCs were cultured on Cytodex type 1 microcarriers in spinner flasks and the characteristics were compared with static culture system. After BM-MSCs
were cultivated and trypsinized from Cytodex type 1 microcarriers, cell viability and proliferation were measured by MTT assay and their phenotypes were analyzed by using fluorescence-activated cell sorter (FACS). These results show that the microcarriers could support the expansion of BM-MSC which can be applied to tissue engineering and regenerative
medicine.
