Acute lung injury is a serious complication of major trauma occurring as a direct consequence of
trauma to the lung or, more commonly, arising indirectly as a consequence of trauma elsewhere to the body. The objective of this study was to scrutinize the involvement of nitric oxide (NO) in
ARDS in a rat model determined by the sequential exposure to lipopolysaccharide (LPS). Rats were injected with 7 mg/kg of endotoxin in pyrogen-free saline. Nitrite measurements in bronchoalveolar lavage fluids (BALF) demonstrated that exposure to microbial products elicits large amounts of NO in LPS challenged rat. LPS infusion led to elevated nitrites level in the BAL. But, injection of taurine and dexemethasone (DEX) alone had effect on the nitrite/nitrate levels in either BAL fluid. Our results show that LPS exposure induces lung damage as demonstrated by protein and lactate dehydrogenase (LDH) increases in BALF. Concomitant taurine and DEX infusion elevated the BAL protein levels; a statistically significant increase compared to rat receiving LPS alone. Measurements of nitrite/nitrate were elevated in the BAL of LPS challenged rats, indicative of an induction of the NO synthase. Inhibition of the NO synthase in LPS challenged mice via the infusion of taurine and DEX led to a very significant increase in nitrites/nitrates in sera and BAL fluids.
