Enhanced digitoxin production in transgenic hairy root cultures of Digitalis purpuria by repeated elicitation

In vitro germinated plantlets of D. purpurea were infected with A. rhizogenes strains (A4, 15834, and 43057) ,which lead to the induction of hairy roots from 10-98% of leaf explants. The highest transformation efficiency of 98% was achieved by using strain 43057 with leaf explants. Polymerase chain reaction analysis confirmed this hairy root lines were of A. rhizogenes origin. Three transformed clones of hairy roots were established. Doubling time of the faster growing hairy root lines was 15 days and these cultures showed about 50-fold increase in biomass at the end of 6 weeks as compared to non-transformed seedling roots. DG-1 root line produced up to 50 ug digitoxin g-1 d.w, which is about 25-fold higher than in in vitro source plants. Methyl jasmonate added at 100 5M on days 14, 21, 28 to cultures of hairy roots growing in flask improved digitoxin production by 56% compared with single addition of 100 5M methyl jasmonate on day 15. The final digitoxin production in the flask reached up to 110 5g digitoxin g-1 d.w ,which is the highest reported for digitoxin production in a flask.
