Stromal-vascular fraction (SVF) is comprised of mixture of cells, such as mesodermal or mesenchymal origin, pericyte, endothelial cells, and smooth muscle cells1). Human adipose derived-SVFs were cultivated in T25 flask up to 3 passages, using DMEM/Ham' s F12(1:1) media with antibiotics at 37∘C under humidified atmosphere of 95% air : 5% CO2. In order to find supplements enhancing SVF proliferation, supplements such as dexamethasone, basic
fibroblast growth factor(bFGF), bovine serum albumin, insulin, transferrin, sodium selenite and fetal bovine serum were tested using human SVF in DMEM/Ham' s F12 media. A cell image analysis software, CellProfiler(www.cellprofiler.org)2), was adopted to trace the morphology change during the growth of SVFs in the presence of various supplements.
Supplementation of bFGF in the DMEM/Ham' s F12 media with serum, significantly increased growth and cell density was increased after 7 days of cultivation, while cell density was 3.0 × 103 cells/cm2 in the media in the absence of bFGF. Cells fixed and stained in the culture plates were analyzed by image analysis software. There was a significant variations for the cell grown
in the presence of bFGF in terms of cell number, area, perimeter, eccentricity, solidity, extent, euler number and form factor.
