Long-term Perfusion Culture of Recombinant CHO cells Producing Recombinant Erythropoietin in Dual Bioreactor System Coupled with a Centritech Lab II Centrifuge

Various parameters such as pH, D.O., temperature and fortified medium was investigated in perfusion culture for 175 days. First of all, low temperature (33℃) was showed significant increase at EPO production 1.35 folds higher than at 37℃ and reduced perfusion rate, simultaneously. Maximum EPO concentration was increased to 2.13 folds. And at the end of this perfusion culture, EPO production at 30℃ was 1.87 folds higher to compared with control. When
fortified medium and 33℃ applied at once, product concentration was increased 1.39 folds higher than low temperature only. Fortified medium only reduced perfusion rate. From 20% to 60% of D.O., perfusion rate was raised to around 3.5 day-1. When pH value of 6.8 was applied, perfusion rate was decreased to 0.77 day-1and EPO was increased to 2.64 folds than control. The produced EPO concentration of 37℃ as control was different in application of various
parameters during perfusion culture, because the cell affected from perfusion culture. So productivity of cell was observed irregular in each case. In this culture, shift to pH 6.8, low temperature and fortified medium (1.5x) with 33℃ were affected to improvement of the EPO production.
