Due to the fact that mycelial cells of S. commune do not sporulate under the submerged cultivation conditions, it is crucial to obtain single cell colonies through protoplast formation for large screening of schizophyllan high-yielding mutants. Schizophyllan, an exopolysaccharide(EPS) produced by submerged cultures of mycelial fungus of Schizophyllum commune has the potent capability of maintaining high moisturized conditions on epidermal tissue, and also having positive effects on the removal of necrotic tissue as well as restoration of epidermal tissue. It was possible to develop a process for rapid and mass screening of high-yielding producers, since large numbers of mutated protoplasts (5x106 protoplasts/ml) could be obtained within 2 hours by applying ST2 method (sonication treatment) with the cell wall disrupting enzymes at a
concentration of 5%. It was found that for the formation of protoplasts, the mycelia should be grown in PL medium at 28℃ for 2 days after homogenization. Under these conditions, the protoplast regeneration rate was as high as 90% when PDA medium containing 1.2M sorbitol was used. Notably, the NTG-mutated strains of S. commune showing high performances in the protoplast regeneration process and rapid growth on solid growth medium were able to stably produce more than 30 g/L of EPS in submerged fermentations. In this paper, we also report that C/N ratio has the most significant influence on the EPS biosynthetic capability of the high-yielding producers.
