Modulation of NIH3T3 Behavior on Mussel Adhesive Protein Fused with RGD Peptide

Previously, we designed and constructed fp-151-RGD by fusing the GRGDSP peptide with the Cterminus of fp-151. Through various cell-adhesion analyses, it was identified that fp-151-RGD had better potentials to be used as a cell or tissue bio-adhesive compared with other commercially produced cell-adhesion materials such as poly-L-lysine (PLL) and Cell-Tak. In this study, fp-151-RGD was prepared and used as a substrate for the in vitro culture of mouse fibroblastic NIH3T3 cells in order to investigate the effect of GRGDSP modification. The fp-151-RGD played a significant role in NIH3T3 cell behaviors. Cell spreading and proliferation rate were remarkably greater than other commercially used cell-adhesion materials. Apoptotic cell rate on fp-151-RGD was also less than any other cell-adhesion materials. Besides, phosphorylated FAK expression level was increased by fp-151-RGD. These results suggest that fp-151-RGD has a positive effect on NIH3T3 and can be used for tissue culture material.
