Solventogenic clostridia are known as one and only bacterial strain capable of production of
butanol by conversion of a variety of carbohydrates. In many other bacteria, clostridia mainly
transport carbohydrates via PEP-dependent phosphotransferase sugar transport system (PTS).
PTS is a group translocation system that consists of more than three enzymes located in both
membrane (Enzyme IIs) and cytosol (HPr and Enzyme I). HPr, a cytosolic component of PTS is
a small regulatory protein involved in carbohydrate transport and in catabolite repression in
gram positive bacteria. In this study, we carried cloning of ptsH gene encoding HPr protein from
C. acetobutylicum and heterologous expression of HPr in E. coli BW 25113 ptsH- , a knockout
mutant of ptsH gene. Both cloning and expression of ptsH from C. acetobutylicum were
performed using either T-vector or pZE21. After transformation of the E. coli mutant, functional
characterization of clostridial HPr protein was carried using a complementation experiment of
PTS sugar utilization such as mannitol. The result confirmed that HPr is a conserved protein
involved among bacteria in terms of the sequence and function.
