Application of engineered bacterial whole mevalonate pathway for β-carotene production in E. coli

The top and bottom portions of MVA pathway from various microorganisms were introduced into recombinant Escherichia coli DH5α harboring β-carotene synthetic genes. The recombinant E. coli with the bottom MVA pathway of Streptococcus pneumoniae produced 141 mg/l of β-carotene for 48 hours of culture with exogenous supplementation of 3.3 mM MVA. It was the highest in comparison with the β-carotene production obtained with the bottom portions of MVA pathway of Enterococcus faecalis, Staphylococcus aureus, Streptococcus pyogenes and Saccharomyces cerevisiae. The production of MVA in the recombinant E. coli harboring the MVA top portion of E. faecalis was 29 mM which was much higher than those of other. The whole MVA pathway was constructed using the bottom and top portions of MVA pathway of S.
pneumoniae and E. faecalis, respectively and introduced into the recombinant E. coli.
