Codonopsis lanceolata, a perennial herb, belongs to Campanulaceae. Codonopsis lanceolata radix has been utilized as a seasonal food and traditional tonic medicine for the past years in korea. Codonopsis lanceolata has been known to be effective for the discharge of phlegm, counteracting poison, asthma, phthisis tuberculosis, etc. This study was concentrated to investigate the identification of anti-allergic effect of various extracts from Codonopis lanceolata. The cascade extraction procedure was used with various solvents from hot-water extract to
obtain several solvents fractions and crude saponin from Codonopsis lanceolata.
Anti-allergic effect of Codonopsis lanceolata based on the production of cytokines and the activation of macrophage was studied. Mast cell releases various mediators upon stimulation. Stimulation of mast cell with compound 48/80 initiates the activation of a signal transduction pathway, leading to histamine release. In pathological skin condition, histamine is involved in the
induction of itching and edema. Therefore, to identify the anti-allergic effect, cytotoxicity was determined by MTT assay. Histamine content was measured using o-phthalaldehyde by spectrofluorometric procedure. NO production was measured using NO assay. Cytokine was measured using western blot.
