ROS Scavenging Activity and Detoxification of Natural Herb against Alcohol-Induced Toxicity and Dysfunction in Liver

The mechanisms responsible for alcohol-induced liver disease are clearly unknown. These effects of ethanol are prominent in liver which is the major site of ethanol metabolism in body. Liver injury is the most common response to alcohol intake, and chronic alcohol consumption generates alcoholic liver disease such as hepatic steatosis, hepatitis and cirrhosis. Alcoholic liver disease results from metabolism confusion and hepatocyte death by ROS. Ethanol
metabolism alters the cytosolic and mitochondrial redox states and disrupts the functions of various metabolic pathways, and promotes oxidative stress both by increasing ROS formation and by decreasing cellular defense mechanisms.
This study evaluated hepatocyte protective effect and reactive oxygen species (ROS) scavenging ability of natural herbs on excessive exposure to alcohol. Natural herbs were extracted by various solvent, and then extraction yield was measured. The cells were cultured at 37℃ in 5% CO₂incubator. After ethanol treatment, hepatocyte was incubated, at once natural herbs were administrated.
Hepatocyte protective effect was evaluated by cell viability through MTT assay and ROS scavenging ability was determined by FACS after fluorescent dyeing.
