To control the quality of the product, photosynthetic bacteria as biomass, by the maintenance of its purity, anaerobic culture and its stabilization were examined. The solid culture on a MYC agar medium under an anaerobic condition using gas pack reactor resulted in maintenance of initial purity (more than 90%) during scale-up from the colony to 1L flask. This culture showed the better purity than that of liquid culture. Some photosynthetic bacteria has been reported to have ability of N2 fixing. Thus, another culture to maintain the purity of broth was experimented on the basal medium eliminating ammonium phosphate and MSG as N-sources, under the presence of N2 gas. This culture also resulted in high purity. Results of cultures on various media indicated the basal medium was much better than LB or TSB medium.
