Major problem in biological H2 production for various applications is low productivity. In order to improve the productivity, immobilization is suggested. In this study, we tried to immobilize Escherichia coli SH5, developed in this lab for H2 production, in different immobilization matrix and studied some parameters to enhance the productivity. Agar as immobilization matrix was good, since floating and breaking of beads due to the production of gaseous H2 and CO2 were not observed at all even though the several times usage for H2 production from formate. To enhance the volumetric production rate, effect of pH, formate concentration and cell concentration were examined. Volumetric production rate increased when pH in acidic pH range and immobilized cell concentration were increased. And 200 mM of formate exhibited the highest volumetric production rate. As a result, more than 2L H2/L/h of volumetric production rate was
obtained under pH 6.5, 200 mM formate and 100 OD cell in beads in this study and this result is more higher than previous reported results.
