Neuronal SNARE (Soluble N-ethylmaleimide sensitive factor attachment receptor) complex
formation between plasma membrane and vesicle membrane is responsible for membrane fusion and neuroexocytosis. Botulinum neurotoxins specifically cleave SNARE proteins resulting in the blocking of neuroexocytosis. This property is now widely harnessed to develop new therapeutic and cosmetic applications. In the present study, we could draw some candidate SNARE folding modulators from phenolic compounds. They acted like Botox by inhibiting the folding of SNARE complex not by cleaving the SNARE proteins. The found phenolic compounds inhibited SNARE complex formation and membrane fusion efficiently. The most potent phenolic compounds, myricetin, delphinidin and cyanidin decreased release of [3H]-noradrenaline from high K+-stimulated PC12 cells. Especially, phenolic compounds had better effect on the inhibition of newly forming SNARE complex than already formed complex. Probably, the phenolic compounds may play a role as an effective antiwrinkle agent like Botox.
