pH-sensitivity of liposome composed of dioleoylphosphatidyl ethanoalmine and cholesteryl hemisuccinate.

pH-sensitive liposomes were prepared by a sonication method using dioleoylphosphatidylethanolamine (DOPE) and cholesteryl hemisuccinate (CHEMS). DOPE and CHEMS were combined for the preparation of the liposomes so that the molar ratios of DOPE to CHEMS are 9/1, 8/2, 6/4 and 5/5. On a transmission electron micrographs, vesicles were observed. The degrees of fluorescence quenching were determined. pH-dependent release properties were evaluated at various pH ranges. Release test in 100 second at low pH, almost 100 % release was observed in 20 sec. When pH increases, we could observe a various release property. Size and zeta potential were measured at various pH conditions.
