Imaging of active targeting using ligand immobilized on the surface modified liposome

For more effective drug delivery and imaging, three properties are considered: target specific delivery, maintenance of drugs' activity, and biocompatibility1). In this study, we characterized EGFR (epidermal growth factor receptor) targeted liposome and imaged how it worked with cell using confocal microscopy. For targeted delivery, we used EGF (epidermal growth factor)
because target cell has EGF receptor on its surface2). In stead of protein drug, Cy5 dyes were entraped in liposomes to image how they work in the cell. To immobilized EGF on the liposome surface, we used BAM (biological anchor for cell membrane) which has oleyl and NHS (N-hydroxysuccineimide)group in each sides and PEG (poly ethylene glycol)between them. EGF was covalently reacted with NHS group, and oleyl group was inserted into the phospholipid of
liposomes. To characterize how BAM immoblized on liposome, QCM (quartz crystal microbalance) was used. Using the lipophilic interaction, it can be inserted to the lipid bilayer to form a plane or a vesicle with the proteins exposed outside. To monitor whether EGF successfully targeted to the cell, we used two cell lines: MCF-7 (EGFR negative) and MDA-231 cell (EGFR positive). After treating EGF decorated Cy5 encapsulated liposome to both
cells, Cy5 signal was selectively monitored in MDA-231 cell.
