Site-specific Cell Behavior from Amnion

Recently some researchers reported that some stem cells exist on and can be isolated from human amnion and amniotic fluid. It is known amniotic epithelial cells (AECs) have some characteristics of stem cells, which could differentiate into ectodermal epidermis, hair follicle and endodermal hepatocyte-like cells, insulin-producing cells. However, the exact origin of the AECs is not clarified in previous reports. So, in this study, we tried to isolate AECs from different
amnion site and confirm whether these different AECs have same characteristics or not. AECs isolated from different amnion site showed same proliferation capacity. However, the isolation success rate was very different from different amnion site; placental < transitional < cord amnion. AECs from different amnion site did not have same characteristics in the point of in situ proliferation and stratification view. However, the isolated AECs had same proliferation and
differentiation (into hepatocyte) capacity. It is not clear why these differences come from, so it will be necessary to investigate their different characteristics and reasons.
