Anti-Wrinkle Activity of Ziyuglycoside I Isolated from Sanguisorba Officinalis Root Extract and Its Application as a Cosmeceutical Ingredient

We previously screened various plants for free radical scavenging activity and inhibition of lastase
activity. Screening showed that Sanguisorba officinalis root extract showed good inhibitory effects against aging. Sanguisorba officinalis is widely distributed throughout Korea and China. Its roots have been used as a traditional oriental medicine for the treatment of internal emorrhage and inflammation.1)-2) In this study, in order to investigate the potential of Sanguisorba officinalis root extract as an active ingredient for wrinkle-care cosmetics, we measured their free radical scavenging activity, elastase inhibitory activity, the expression of MMP-1 (matrix etalloprotease-1) in vitro, and type I collagen synthesis in normal human fibroblast cells. Sanguisorba officinalis root extract (SC50 = 7.4 Bg/ml) showed very high free radical scavenging activity compared to BHT (di-t-butyl hydroxytoluene) (SC50 = 25.4 Bg/ml), which was used as a positive control. Elastase inhibitory activity of Sanguisorba officinalis root extract (IC50 = 43.0 Bg/ml) was higher than that of oleanolic acid (IC50 = 83.8 Bg/ml). Sanguisorba officinalis root extract reduced the expression of MMP-1 (up to 40.6% at 50 Bg/ml). Sanguisorba officinalis root extract with superior effects on elastase activity inhibition also showed high DPPH radical scavenging activity. This
result suggested that Sanguisorba officinalis root extracts have potential as anti-wrinkle agents when used as ingredients of cosmetics. In this work, to isolate the main component compounds from Sanguisorba officinalis root extract, we purified the extract through solvent fractionation, column chromatography, and recrystallization. It was identified as Ziyuglycoside I using pectroscopic analysis.3) Ziyuglycoside I increased the expression of type I collagen in a dose-dependent manner (up to 71.3% at 50 BM), comparable to that of ascorbic acid (up to75.2% at 250 BM). Ziyuglycoside I showed no cytotoxicity up to the effective concentration for anti-wrinkle activity (less than 50 BM). The 20 subjects (35~53 years female in good general health) were recruited for the clinical study of the formulation, contained Ziyuglycoside I. This double-blind, placebo-controlled, left-right randomized clinical study was carried out during a 12-week to assess the test and placebo formulation (the test cream contained 0.03% Ziyuglycoside I and placebo does not).4) In this clinical test, we measured its efficacy in anti-wrinkle effects on the skin by the visual evaluation of dermatologists, photometric evaluation, manufacture of skin replicas and image analysis using the Skin-Visiometer SV 600. In an image analysis of skin replicas, the test formulation showed more improvement in the average roughness difference (ΔR3, -0.03 ± 0.02) than those of placebo formulation (ΔR3; -0.01 ± 0.02). In the clinical study of measurements using visual evaluation and image analysis showed a statistically significant difference (p<0.05) between the effects of the test and placebo products. This result suggests that Ziyuglycoside I isolated from Sanguisorba officinalis root extract could be used as an active ingredient for new anti-wrinkle cosmetics.
