Effects of Acetates on Cell Viability and hCTLA4Ig Production in

A transgenic Oryza saitva L. cell line was used to produce hCTLA4Ig with RAmy3D promoter.1) This promoter is induced strongly when sugar is exhausted in culture media. Although RAmy3D promoter system enables transgenic rice cells to express recombinant proteins highly, the cells in the phase of induction cannot maintain their viability because there are no energy sources in culture media. Therefore, it is necessary to supply another energy source which does not suppress the promoter with the maintenance of cell viability. It has been reported that the rice cells used acetate in advance as a carbon source when glucose and acetate were mixed in the culture media.2,3) This study investigated the effects of acetates as an alternative energy source
on the cell viability as well as the production of hCTLA4Ig in transgenic rice cell suspension cultures.
