Strain improvement and statistical medium optimization were performed for enhanced production of β-D-glucan in submerged mycelial cultures of Sparassis crispa. β-D-glucan biosynthesized by S.crispa has been well-known to have strong immunostimulating characteristics. Optimization of the production medium was performed systematically using the powerful statistical experimental designs such as Plackett-Bueman design, fractional factorial design(FFD). steepest ascent method(SAM) followed by response surface method(RSM). Notably, it was observed that the produces morphology should be in the form of compacted pellet for the maximum production of β-D-glucan in the production fermentations, the diameter of which should be in the range between 0.5 mm and 1.0 mm. In fact, almost all the screened mutants with high productivities showed compact-pelleted morphologies in the production cultures. In addition to the good morphological characteristics, the components and compositions of the
production medium were observed to be very important for the maximal utilization of the producers' fermentation physiology leading to active biosynthesis of β-D-glucan.
