The fish Cytochrome P450 1A (CYP1A) gene is a useful biomarker in assessing contamination of the aquatic environment1, 2, 3. Induction of cytochrome P450 1A gene expression has been used as a biomarker for aromatic and organic compounds exposure in teleost fishes. We cloned and sequenced full-length cDNA of hepatic CYP 1A gene from river puffer fish, Takifugu obscurus. The open reading frame of T. obscurus CYP 1A cDNA was observed to be 1566 bp that encoded 521 amino acid of putative protein, showing 5.74 of theoretical pI and 59.2 kDa of Molecular weight. By pfam domain searching, we found p450 domain on the position 45 – 513 aa. T. obscurus CYP1A amino acid sequences shared high identity with yellow puffer (Tetraodon nigroviridis ,82%), Marble sole (Pleuronectes yokohamae, 81%) and European sea bass (Dicentrarchus labrax, 80%) in the result of blast search. Changes of CYP 1A gene expression was measured in eight tissues (brain, eye, gill, gonad, heart, intestine, kidney and liver) using real-time RT-PCR after exposure to PAH-like compound, beta-naphthoflavone (BNF) (50mg/kg) for 96h. T. obscurus CYP1A transcripts were ubiquitously distributed in all examined tissues. BNF-induction of CYP1A mRNA expression was detected in most of examined tissues except for gill and heart. These results suggest that CYP1A gene in T. obscurus involve in detoxification of BNF. Furthermore, our findings indicate that T. obscurus CYP1A gene expression would prove to be a useful biomarker of exposure to BNF.
