Regulation of a Cold Shock Protein(csp) Gene by External Stresses in Bifidobacterium breve KB 69

Bifidobacterium species are Gram-positive, anaerobic branched of pleomorphic rods, and one of the normal flora in the large intestine of human and animals. They play and important role in maintaining health by producing antimicrobial substances or bacteriocins to inhibit undesirable pathogens.1) During food products with bifidobacteria produce cold shock protein to enhance or maintain their viability. In this study, csp gene was isolated from Bifidobacterium breve KB 69 by degenerate PCR and single primer PCR technique. The regulation of csp gene was investigated under various stressed at transcriptional level. The amount of csp mRNA was measured by using reverse transcriptase polymerase chain reaction. The study of the cold shock response d adaptation is important for our understanding of how microorganisms are able to live under
low temperature stress and for improvement of cold tolerance in useful microbes. The study is also important in bacteria such as Bifidobacterium spp. and Lactobacillus spp., which are widely used in the dairy industry. Bifidobacterium breve , cold stress, cold shock protein (CSP)
