Production & Analysis of Recombinant Mussel Adhesive Protein FP-151 in insect Sf9/baculovirus Expression System

Mussel adhesive protein (MAP) has strong adhesion with various substrates even in wet environment. Moreover MAP is environmental friendly. In this study, we constructed an insect Sf9/baculovirus expression system for MAP production. Because Baculovirus can efficiently infect insect cells, it is more adaptable system for eukaryotic gene expression. Baculovirus replication level is high when host cell is insect Spodoptera frugiperda CLONE9 (Sf9) cells. Thus, we can efficiently increase expression level through amplification. In order to develop the recombinant baculovirus system, we obtain the MAP gene from foot of Mytilus galloprovincialis and add 6xHis-tag for protein purification. We demonstrated recombinant MAP was successfully expressed in infected Sf9 cells. We studied cell attachment and Serine phosphorylation analysis.
