Proteins Associated to Astaxanthin Accumulation by Green Alga Haematococcus lacustris (Chlorophyceae) Identified using Proteomics Analysis

The unicellular green alga Haematococcus lacustris (Chlorophyceae) is known for the
ketocarotenoid astaxanthin (3,3’-dihydroxy-β,β-carotene-4,4’-dione) accumulation, which is
induced under unfavorable culture conditions such as high irradiance, high temperature, nutrient
deficiency, and oxidative stress from reactive oxygen species (ROS). In this study, H. lacustris
cells were cultivated in photobioreactor columns and induced to accumulate astaxanthin
production by exposure to continuous light intensity after the cells grew into stationary phase and
nitrogen-depleted condition. Using 1-D, 2-D electrophoresis comparison, the differently-expressed
proteins of various cell states according to induction time were selected for further analysis by
matrix assisted laser desorption / ionization time-of-flight mass spectrometry (MALDI-TOF-MS)
to identify the protein functions. Proteins associated to astaxanthin biosynthesis pathway of H.
lacustris were identified and their expression patterns were analyzed.
