A unicellular green alga, Haematococcus pluvialis, has been focused as a microbial source of astaxanthin. In this study, we use Haematococcus pluvialis SC1 and SC2 which are resistant of nicotine by random mutation treated by methanesulfonic acid. Astaxanthin is a high-value antioxidant fifty times stronger than β-carotene, as a food supplement for humans has been suggested. Haematococcus pluvialis photoautotrophic induction has been two-stage. The
first stage was cultivated cell with NIES-C medium, 200 μM nicotine and 5% excess CO2 gases. The second stage, we change medium from NIES-C to NIES-N and provide high light because it is good to be a rapid induction. We investigated for enhancement of astaxanthin production using initial N concentration change (0 mM NO3, 1 mM NO3, 2 mM NO3, 4 mM NO3) in photoautotrophic induction condition.
